Graft-versus-host disease in rats after donor treatment with cyclophosphamide and spleen cells of host origin.

چکیده
Lethally irradiated Lewis-Brown Norway F1 hybrid (LBN) rats were grafted with either marrow or marrow and spleen cells from normal Lewis (L) donors. All recipients of inocula containing both marrow and spleen cells died of graft versus host (GVH) disease. In other irradiated and grafted rats, the severity of the GVH disease was reduced by challenge of the donor with LBN spleen cells prior to or after treatment with cyclophosphamide (CY). The magnitude of this suppressed immunity depended on the interval between the administration of CY and the ailogeneic spleen cells. Maximum suppression was obtained when donors received CY on day 0 and LBN cells on day +1. In all experiments, the treated donors were sacrificed on day +7. The observed immunosuppression was shown to result from the synergistic action of CY and cells bearing the alloantigens of the recipient. Furthermore, the phenomenon was demonstrated to be antigen specific.

Immunochemotherapy of transplantable Moloney leukemia with cyclophosphamide and allogeneic spleen lymphocytes and reversal of graft-versus-host disease with alloantiserum.
Histoincompatible (H-2) spleen cells from C57BL/6 mice that were sensitized with Moloney sarcoma virus caused fatal graft-versus-host disease in BALB/c mice when the cells were used in conjunction with an immunosuppressive, chemotherapeutic dose of cyclophosphamide to treat transplantable Moloney virus-induced leukemia. When antiserum against the donor spleen cells was administered 2 days after...
متن کاملRenal cancer treatment with low levels of mixed chimerism induced by nonmyeloablative regimen using cyclophosphamide in mice.
Recently, much attention has been paid to nonmyeloablative allogeneic stem cell transplantation for the treatment of metastatic renal cancer. Mature donor T cells cause graft-versus-host disease (GVHD) although they are also the main mediators of the beneficial graft-versus-tumor activity associated with this treatment. Hence, the segregation of the graft-versus-tumor activity from GVHD is an i...

Background: Graft-versus-host disease is one of the major complications after allogenic bone marrow transplantation, but it is not easy to anticipate the onset. Cytokines released by type 1 T-helper cells are thought to play a pivotal role in acute graft-versus-host disease (aGVHD). The ability to predict the likely occurrence of graft-versus-host-disease (GVHD) after BMT would be extremely val...
متن کاملRelationship between Molecular Chimerism and Graft Versus Host Disease after Allogenic Hematopoietic Stem Cell Transplantation
Background: Bone marrow transplantation (BMT) is considered as a curative therapy for a broad range of diseases. However, complications such as relapse and graft versus host disease (GVHD) may be observed following BMT. Chimerism analysis serves as a reliable indicator of transplant outcome. Complete chimerism refers to the complete replacement of hematopoietic system by donor cells, while mixe...
